Gradient O2 within the individual wells as the bottom of the samples (cells) experience the higher O2 concentration through the gas permeable membrane and the elevated O2 chamber while the top of the transwell is exposed to the anaerobic conditions of the chamber.

Unlike other systems the Coy Co-culture system allows direct contact between bacterial and mammalian cells. And unlike the microfluidic systems or solutions using a transwell it is far simpler and less labor-intensive to operate.

  • Well-established plasma surface modification includes amination.

  • Suitable for growing most adherent cells (e.g. HEK U293, HepG2, MCF-7)

  • Coating with extra cellular matrix proteins is recommended for some very sensitive cell lines (e.g. PC-12) and some primary cellsrimary cells.

The system consists of:

  • Small chamber
  • Gas permeable plate
  • Contained within an Anaerobic Chamber
  • Tubing and flow meters connect the system to a specific oxygen tank

The Chamber allows for a gas permeable well plate to be exposed to elevated Oxygen levels while maintaining anaerobic conditions in the surrounding glove box. The results are a uniform stratification of the media based on media depth and location to the top of the well (anaerobic) or bottom (desired microaerophilic levels).

Gas Permeable Plate is sealed to the chamber below through gas tight seals, top and thumb screws. Quick disconnect fittings make tubing connection quick and easy. Set up in less than 10 minutes.

In testing O2 concentrations have been tested through a variety of dissolved O2 sensors at consistent and defined positions within the Transwell with the use of a micromanipulator.

The Coy Co-Culture system allows users to use any anaerobic chamber with minor modifications for tubing to be used.
